Nicotinamide adenine dinucleotide (NAD⁺) is an endogenous pyridine nucleotide coenzyme that functions as an obligate hydride-transfer intermediate in over 500 enzymatic reactions. As the oxidised form of the NAD⁺/NADH redox couple, it participates in glycolysis, the citric acid cycle, and mitochondrial oxidative phosphorylation. Beyond its canonical role in redox metabolism, NAD⁺ is consumed as a substrate by sirtuin deacylases (SIRT1–7), poly(ADP-ribose) polymerases (PARPs 1–3), and CD38/CD157 ectoenzymes, linking cellular energetics to epigenetic regulation and DNA damage response pathways. Research interest has expanded substantially following characterisation of the Preiss-Handler, de novo, and salvage biosynthetic pathways and their differential tissue expression.
