Retatrutide (LY3437943) is a synthetic polypeptide investigational compound developed by Eli Lilly and Company, functioning as a triple agonist at glucagon-like peptide-1 receptor (GLP-1R), glucose-dependent insulinotropic polypeptide receptor (GIPR), and glucagon receptor (GCGR). Co-agonism at all three incretin-related receptors is hypothesised to elicit additive or synergistic effects on energy homeostasis and lipid metabolism in preclinical in vitro assay systems. Phase II clinical trial data (NCT05582538, results published 2023 in the New England Journal of Medicine) documented changes in relevant metabolic biomarkers in enrolled human subjects; however, all mechanistic in vitro findings should be interpreted independently of clinical outcomes and do not constitute evidence of efficacy for any application.
